چکیده

Adipocyte fatty acid binding protein, aP2, present in macrophages has been implicated in the integration of lipid metabolism and inflammatory response, contributing to development of insulin resistance and atherosclerosis. We investigated the modulation of aP2 expression by inflammatory insults and fatty acids in RAW 264.7 macrophages. When the cells were incubated with lipopolysaccharides (LPS; 100 ng/ml) or 10 ng/ml of tumor necrosis factor α for 18 h, aP2 mRNA and protein levels were drastically increased. BSA), but not saturated fatty acids (16:0), significantly repressed the basal aP2 expression and abolished induction of aP2 expression by LPS. Trichostatin A (TSA), a histone deacetylase (HDAC) inhibitor, increased aP2 mRNA levels but abolished the repressive effect of 18:2 on aP2 expression in unstimulated and LPS-stimulated macrophages. Cells transfected with siRNA targeting aP2 showed a relationship with cyclooxygenase-2 (COX-2), but not other pro-inflammatory mediators. In summary, our data suggest that unsaturated fatty acids may inhibit the basal as well as LPS-induced aP2 expression by mechanisms involving HDAC in RAW 264.7 macrophages and that there may be a link between COX-2 and aP2-repression by unsaturated fatty acids.

Effects of feed restriction and dietary fat type on mRNA expression of liver fatty acid-binding protein (L-FABP) in broilers

Background: Liver fatty acid-binding protein (L-FABP) is the main cytosolic binding site for long chain fatty acids in hepatocytes. FABPs enhance the uptake of fatty acids into the cell by increasing their concentration due to decreasing concentration of unbound fatty acids inside the cell. Objectives: The aim of this study was to evaluate the effects of dietary unsaturated to saturated fatty a...

Compare the Effect of Eicosapentaenoic Acid and Oxidized Low-Density Lipoprotein on the Expression of CD36 and Peroxisome Proliferator-Activated Receptor Gamma

Background: There is evidence that CD36 promotes foam cell formation through internalizing oxidized LDL (ox-LDL) into macrophages therefore, it plays a key role in pathogenesis of atherosclerosis. In addition, CD36 expression seems to be mediated by nuclear receptor peroxisome proliferator-activated receptor gamma (PPAR-γ). The aim of the present study was to evaluate and compare the effect of ...

تعیین اثر اسید الاییدیک بر بیان ژن PPARγ در رده سلولی ماکروفاژی RAW 264.7

Background: Several dietary factors are involved in cardiovascular coronary heart diseases, including trans fatty acids, which are generally formed during hydrogenation of vegetable oils, a process that causes conversion of liquid oils into semisolid fats. Nowadays, it is well-known that trans fatty acids form a major risk factor in the occurrence and progression of atherosclerosis. On the ot...

Regulation of adipose cell differentiation. II. Kinetics of induction of the aP2 gene by fatty acids and modulation by dexamethasone.

Fatty acids behave as activators of the aP2 gene expression in committed, lipid-free, non-terminally differentiated Ob1771 cells. Like fatty acids, dexamethasone provokes a dose-dependent accumulation of aP2 mRNA. However, fatty acids and dexamethasone act through different mechanisms to activate the aP2 gene expression since i) fatty acids and dexamethasone act in a synergistic manner; ii) the...
